RESUMO

In the current study, an extracellular cellulase belonging to symbiotic Bacillus subtilis Bc1 of the leopard moth is purified and characterized. The molecular mass of enzyme was 47.8 kDa using SDS-PAGE. The purified enzyme had optimum activity in temperature and pH around 60 °C and 8, respectively. The purified cellulase was introduced as a stable enzyme in a wide variety of temperature (20-80 °C) and pH (4-10) and remained active to more than 74% at 80 °C for 1 h. Moreover, the cellulase extremely was stabled in the presence of metal ions and organic solvents and its activity was increased by acetone (20% v/v), CaCl2 and CoCl2 and inhibited by MnCl2 and NiCl2. The values of enzyme's Km and Vmax were found to be 1.243 mg/mL and 271.3 µg/mL/min, respectively. The purified cellulase hydrolyzed cellulose, avicel and carboxymethyl cellulose (CMC) and the final product of CMC hydrolysis was cellobiose using thin-layer chromatography analysis. Consequently, owing to exo/endoglucanase activity and organic solvent, temperature and pH stability of the purified cellulase belong to B. subtilis BC1, it can be properly employed for various industrial purposes.

The present study aimed to explore some simple ways to optimize the mass rearing of Cotesia vestalis (Haliday) (Hymenoptera: Braconidae), one of the most important biocontrol agents of the diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae). The effects of host and parasitoid initial densities on the quantity and quality of produced wasps were investigated. In particular, a factorial experiment was established using three different host densities (20, 50, and 100 P. xylostella larvae per cage) in presence of C. vestalis at three different densities (2, 5, and 7 females, with a 24-h exposure) in well-ventilated cages. There was a significant effect of wasp's initial density on the percentage parasitism (measured based on the cocoon and emerged adult wasps); comparatively the greatest percentage parasitism was obtained with five wasps per cage. Initial host density had no significant effects on the percentage parasitism, sex ratio, and larval period, but it influenced the survival rate (measured based on the cocoon and adult emerged) and pupal period of produced wasps, and also percentage host mortality. The density of 20 host larvae was the best in this regard. These findings were discussed in the contexts of optimum mass rearing, life history, and potential of C. vestalis as a biological control agent.

RESUMO

The Iranian checklist of the tribe Chilocorini Mulsant, 1846 (Coleoptera: Coccinellidae) is updated. In total, 13 species belonging to four genera (Brumoides Chapin, 1965, Chilocorus Leach, 1815, Exochomus Redtenbacher, 1843, and Parexochomus Barovsky, 1922) are listed from Iran. An identification key to all genera and species currently known from Iran is presented along with illustrations of adult specimens and male genitalia.

RESUMO

The Iranian species of the tribe Hyperaspidini Mulsant, 1846 (Coleoptera: Coccinellidae) are reviewed. The current list includes 12 species, all placed in a single genus Hyperaspis Chevrolat, 1836. Hyperapsis asiatica Lewis, 1896 and H. pumila Mulsant, 1850 are excluded from the Iranian list of Coccinellidae. Diagnoses of the tribe Hyperaspidini and the genus Hyperaspis are given. Images of adult beetles and diagnostic characters of the male genitalia of all species distributed in Iran are shown. A key to identification of the species is presented. Distribution records are provided for each species along with information on host plants and prey species when available.

RESUMO

An updated checklist of the Coccinellidae species of the former subfamily Sticholotidinae recorded from Iran is provided. Eleven species are reported: two species classified presently in the subfamily Microweiseinae (in the genera Paracoelopterus Normand, 1936 and Serangium Blackburn, 1889), and nine species classified in the tribe Sticholotidini of the subfamily Coccinellinae (in the genera Coelopterus Mulsant & Rey, 1852 and Pharoscymnus Bedel, 1906). Pharoscymnus smirnovi Dobzhansky, 1927 is removed from the list of the Coccinellidae of Iran. Distribution of species in Iranian provinces is presented. Data concerning their host plants along with their prey species are also included when known. Morphological features of two rarely collected and poorly known species of Iranian fauna, Pharoscymnus brunneosignatus Mader, 1949 and Pharoscymnus pharoides (Marseul, 1868) are diagnosed and illustrated.
